You can certainly bill for the colposcopy of the vagina using 57420, and I assume they are removing vaginal mucosa lesions with the laser. As there is no code for this type of procedure specifically you might consider that 57135, excision of vaginal tumors would be close to the work done. So perhaps bill 57420-22 or 57420 and 58999 (unlisted) and compare the additional work to 57135.
