If a patient comes into the office for a vaccination and for what ever reason leaves before it is administrated but it has already been drawn, is the doctor able to charge patient or the patients insurance?

A 53 would not be appropriate in this case. You could only code for the vaccine product. The documentation would need to include the information regarding the prep completion and the patient leaving to indicate why no administration code is being billed.
